just if one uses the hot shoe or TTL connector?

Yes. Rearranging the 1st equation, you get.
marked f# = (Effective f#) / (m+1)
The effective f# is what the film will experience.
The marked f# is the lens marking.
Diffraction goes by the effective f#, so it's best to not go past a
value of f/16.
