You perhaps missed my post of a few days ago pointing to a free DOF and
hyperfocal distance calculator from Digital Light & Color, publisher of
Picture Window.
Go to:

and click on downloads at the left of the page and then scroll down
looking for the DOF calculator.
From the required input parameters you will see that DOF is controlled
only by focal length, f stop, focus distance from subject and the
desired "circle of confusion", here represented by resolution in
lines/mm. In other words, how sharp does it have to be to be considered
in focus.
What's in or out of focus gets a bit tricky in that something you might
deem to be in acceptable focus on a 4x6" print might look out of focus
on an 8x12" print. So, the final magnification of the image is a
consideration in the DOF caclculation which you would control by setting
the required resolution.
Unfortunately, the 50mm f/3.5 as a high quality macro lens can't give us
any better depth of field than any other 50mm lens. It will, however,
probably have a flatter field and be sharper than many other lenses
which might influence the appearance of DOF.
