This is the advice from the Reference Manual (electronic version):
Focusing during live view
• When you press the shutter button all the way
during live view, the focus is locked and a picture
is taken. The image display on the monitor
freezes as the mirror goes down during focusing.
• To focus in advance, hold down the AEL/AFL
button and press the shutter button. When the
focus is locked, the AF confirmation mark lights
up. When the AF confirmation mark is blinking,
the focus is not locked.
• When the AF mode is set to S-AF, the picture
cannot be taken until the focus is locked.
• In MF mode, you can press the AEL/AFL button
to activate the auto focus.
